產(chǎn)品介紹 Streptavidin is a tetrameric protein composed of identic subunits. Each subunit binds one biotin molecule with a KD of ~1x10-15M. The preparation contains an N- and C-terminal shortened variant (core streptavidin) with improved properties concerning homogeneity, solubility, resistance towards proteolytic degradation and accessibility of the biotin binding pocket as compared to native streptavidin. The high affinity recognition of biotin and biotinylated molecules has made streptavidin one of the most important components in diagnostics and laboratory kits.

鏈霉親和素具有與親和素(Avidin,AV)相似的生物學(xué)特性,如都能與4分子的生物素(Biotin)特異性結(jié)合,結(jié)合常數(shù)為1×10-15/M。同時(shí),由于SA不帶糖基、等電點(diǎn)低,在檢測(cè)中具有比AV低的背景而大大提高了檢測(cè)的靈敏性。
生物素與親和素/鏈霉親和素的結(jié)合具有高度特異性和穩(wěn)定性,兩者的親和常數(shù)比抗原-抗體反應(yīng)高10-100萬(wàn)倍,是目前已知強(qiáng)度最高的非共價(jià)作用,這使得它成為免疫學(xué)檢測(cè)中的一個(gè)非常好信號(hào)放大的工具。廣泛應(yīng)用于酶聯(lián)免疫反應(yīng)、熒光分子標(biāo)記免疫反應(yīng)和分子雜交為原理的各種檢測(cè)試劑盒的SA-biotin系統(tǒng)。但目前,國(guó)內(nèi)所用SA均為價(jià)格昂貴的進(jìn)口產(chǎn)品,因此研制和生產(chǎn)國(guó)產(chǎn)化的SA具有重要的現(xiàn)實(shí)意義和廣闊的應(yīng)用前景。
????鏈霉親和素是與親和素具有相似性質(zhì)的一種蛋白質(zhì),都具有與生物素特異結(jié)合的能力,結(jié)合常數(shù)高達(dá)1015M。同時(shí),由于鏈霉親和素等電點(diǎn)比親和素低,且不含糖基鏈,故在檢測(cè)中的靈敏度和特異性都高于親和素,建立在此基礎(chǔ)上的生物素-鏈霉親和素系統(tǒng)的應(yīng)用比生物素-親和素系統(tǒng)有更大優(yōu)勢(shì)。
????我公司生產(chǎn)用鏈霉親合素-SA菌株及生產(chǎn)工藝均自國(guó)外原本帶回。生產(chǎn)工藝穩(wěn)定,保證質(zhì)量,常年提供產(chǎn)品,確保供應(yīng)。
我公司生產(chǎn)的SA單體經(jīng)SDS-PAGE檢驗(yàn)分子量為17.2KDa。經(jīng)論證,完整的SA溶解性差、易于聚集,核心SA被蛋白酶水解掉了與活性無(wú)關(guān)的某些末端序列,一方面提高了結(jié)構(gòu)的穩(wěn)定性;另一方面由于減少了空間位阻,使SA更容易與生物素標(biāo)記的大分子發(fā)生反應(yīng)。
SA的純化是用2-亞氨基生物素瓊脂糖珠4B 凝膠(Sigma 產(chǎn)品)進(jìn)行親和層析純化,同時(shí)以離子交換進(jìn)行進(jìn)一步的精細(xì)純化。
SA分子量為58KDa,生物活性15.7U/mg,接近理論值(16.8U/mg)并進(jìn)行了活性測(cè)定。
SA溶解與保存:我公司提供的SA產(chǎn)品,不含任何鹽離子,用戶可以根據(jù)自己的試驗(yàn)需要,使用不同的緩沖液進(jìn)行溶解,溶解后應(yīng)放與-20°C保存,避免反復(fù)凍溶。
